Macrocybin,a Natural Mushroom Triglyceride,Reduces Tumor Growth In Vitro and In Vivo through Caveolin-Mediated Interference with the Actin Cytoskeleton

Mushrooms have been used for millennia as cancer remedies. Our goal was to screen several mushroom species from the rainforests of Costa Rica, looking for new antitumor molecules. Mushroom extracts were screened using two human cell lines: A549 (lung adenocarcinoma) and NL20 (immortalized normal lung epithelium). Extracts able to kill tumor cells while preserving non-tumor cells were considered “anticancer”. The mushroom with better properties was Macrocybe titans. Positive extracts were fractionated further and tested for biological activity on the cell lines. The chemical structure of the active compound was partially elucidated through nuclear magnetic resonance, mass spectrometry, and other ancillary techniques. Chemical analysis showed that the active molecule was a triglyceride containing oleic acid, palmitic acid, and a more complex fatty acid with two double bonds. The synthesis of all possible triglycerides and biological testing identified the natural compound, which was named Macrocybin. A xenograft study showed that Macrocybin significantly reduces A549 tumor growth. In addition, Macrocybin treatment resulted in the upregulation of Caveolin-1 expression and the disassembly of the actin cytoskeleton in tumor cells (but not in normal cells). In conclusion, we have shown that Macrocybin constitutes a new biologically active compound that may be taken into consideration for cancer treatment.     

开放式复杂航空网络系统的动力学演化

本文首先实证研究了中国航空网(CAN)与外部经济环境,即国民生产总值(GDP)的相关性,进而揭示CAN的演化方式及其拓扑特征的起源.发现自1998年起网络节点的度与其GDP成线性关系,表明了网络拓扑与经济因素有密切的联系,且CAN的度演化服从类似于多重过程(multiplicative process)的模式,而该模式又强烈依赖于GDP的增长率.这种独特的模式暗示了CAN具有类似适应度模型的微观机理.通过对演化方程的研究发现:1)CAN在增长过程中,节点加入的时刻具有经济上的偏好性,即节点加入的时刻与该节 关键词： 国民生产总值(GDP) 中国航空网(CAN) 双段幂率分布 多重过程     

蛋白质对测定水产品中氯苯类化合物的影响及消除

提出了气相色谱-质谱法测定水产品中1,3-二氯苯,1,2-二氯苯,1,2,4-三氯苯等3种氯苯类化合物含量的方法。采用荧光猝灭法研究了氯苯类化合物与蛋白质之间的结合作用,结果表明氯苯类化合物与鱼血清白蛋白之间存在较强的结合作用。水产品样品以丙酮-磷酸氢二钾-水双水相体系进行前处理。在气相色谱分离中用DB-WAX毛细管色谱柱为固定相,在质谱分析中采用选择离子监测模式。3种氯苯类化合物的质量浓度均在0.5~5.0mg·L-1范围内与其峰面积呈线性关系。以空白样品为基体进行加标回收试验,所得回收率在77.0%~116%之间,相对标准偏差(n=6)在2.6%~7.1%之间。     

Fractional pyrolysis of Cyanobacteria from water blooms over HZSM-5 for high quality bio-oil production

Fractional pyrolysis and one-step pyrolysis of natural algae Cyanobacteria from Taihu Lake were comparatively studied from 200 to 500 ℃. One-step pyrolysis produced bio-oil with complex composition and low high heating value （HHV〈30.9 MJ/kg）. Fractional pyrolysis separated the degradation of different components in Cyanobacteria and improved the selectivity to products in bio-oil. That is, acids at 200 ℃, amides and acids at 300 ℃, phenols and nitriles at 400 ℃, and phenols at 500 ℃, were got as main products, respectively. HZSM-5 could promote the dehydration, cracking and aromatization of pyrolytic intermediates in fractional pyrolysis. At optimal HZSM-5 catalyst dosage of 1.0 g, the selectivity to products and the quality of bio-oil were improved obviously. The main products in bio-oil changed to nitriles （47.2%） at 300 ℃, indoles （51.3%） and phenols （36.3%） at 400 ℃. The oxygen content was reduced to 7.2 wt% and 9.4 wt%, and the HHV was raised to 38.1 and 37.3 MJ/kg at 300 and 400 ℃, respectively. Fractional catalytic pyrolysis was proposed to be an efficient method not only to provide a potential solution for alleviating environmental pressure from water blooms, but also to improve the selectivity to products and obtain high quality bio-oil.     

有机化学进展(2011~2012)

本文综述了中国大陆地区有机化学研究人员2011至2012年两年内在合成方法学、有机合成化学、元素有机化学以及天然产物化学等领域获得的重要成果。文章中共引用参考文献355篇,其中110多篇手性金属配合物和有机小分子催化的不对称反应、金属催化的碳氢键活化等合成方法学论文和30余篇氟有机化学论文基本来源于德国《应用化学》(国际版)和《美国化学杂志》。本文汇集了中国有机化学家两年中合成的150多个具有生物活性和化学结构多样性的天然产物,其中不乏具有高度挑战性的复杂天然分子。在近两年中中国有机化学家从陆地和海洋的生物体内发现各种不同类型新天然产物90多个。     

A Solvent Effect in the Reaction of Diazomethane with Norbornane-2,3-dione 3-Hemiketals

A solvent effect in the reaction of diazomethane with norbornane-2,3-dione 3-hemiketal in various aprotic solvents such as ether, dichloromethane, tetrahydrofuran (THF), dioxane, and chloroform is reported. Unlike in methanol, preparatively useful quantities of oxetane derivatives were obtained along with a novel hexahydro-1H-cyclopenta[c]furan-1-one in these aprotic solvents. Dichloromethane and THF gave optimal yields of the two products. In some cases, α-ketoketals were formed as minor products along with the aforementioned products. Interestingly, TMSCHN₂ furnished hexahydro-1H-cyclopenta[c]furan-1-one as the predominant product.     

Recent advances of desymmetrization protocol applied in natural product total synthesis

Desymmetrization synthesis strategy has simplified and improved the efficiency of synthesis, which attracted great attention in the past few decades. Since the strategy has been developed rapidly and got a wide range of applications in natural product total synthesis, the rules are urgent to be summarized. In this Letter, the recent developments of desymmetrization protocol in natural product total synthesis were summarized.     

Formal and total synthesis of (±)-cycloclavine

A ring fragmentation and intramolecular azomethine ylide 1,3-dipolar cycloaddition sequence of reactions was successfully used in the preparation of a known (±)-cycloclavine precursor in good overall yield. Results of efforts to incorporate the tetrasubstituted cyclopropane ring present in cycloclavine are also discussed.     

An enantioselective synthesis of a MEM-protected aetheramide A derivative

Aetheramides A and B are very potent anti-HIV agents. An enantioselective synthesis of a MEM-protected aetheramide A derivative is described. The synthesis was accomplished in a convergent and stereoselective manner. The key reactions involved asymmetric dihydroxylation, asymmetric allylation, asymmetric syn-aldol reactions, and asymmetric hydrogenation.     

Effect of temperature on CO2 corrosion of carbon steel

This work investigates the effect of temperature on the corrosion product layer of carbon steel exposed to a CO₂‐containing solution. The measurement techniques, such as scanning electron microscopy with energy dispersive spectrometry, X‐ray diffraction, and X‐ray photoelectron spectroscopy, were used to systematically characterize the morphology and composition of the corrosion product layer. The corrosion rates were calculated by weight loss method. The corrosion mechanisms as a function of temperature are studied and discussed. The results showed that temperature is an important factor in the corrosion rate of carbon steel. Copyright © 2009 John Wiley & Sons, Ltd.
